1株登革Ⅰ型病毒的分离鉴定及系统进化分析

Isolation, identification and phylogenetic analysis of a dengue virus I

摘要: 目的 了解1株登革病毒分离株的来源及进化情况。方法 分离获得1株登革病毒,通过对该毒株进行血清型分型鉴定及基因组序列测定,进而通过生物信息学方法对囊膜蛋白E进行系统进化分析。结果 分离获得1株DENV血清Ⅰ型基因Ⅰ型毒株;且E基因的同源性分析显示,与Malaysia 0802aTw、Singapore株、浙江17-45株、YN176282株、BNJH17108株的核苷酸同源性均在99%以上;氨基酸水平,与NJX-14株、浙江17-45株、Malaysia 0802aTw株、1208aTw株等多种毒株的E蛋白的同源性均为99.8%, 表明这些毒株可能源于同一株原始毒株,且E蛋白序列遗传进化稳定,与2002年以来多株广东省分离的毒株高度同源。结论 近十年发现在我国的DENVⅠ型基因Ⅰ型遗传进化稳定,保守程度高,有利于针对该血清型登革病毒的防控。

Abstract: Objective To preliminarily understand the origin and evolution of the dengue virus isolated strain. Methods A dengue virus was isolated, and its serotyping and genome sequence was identified and analyzed. Especially, the phylogenetic analysis of envelope protein E was performed by bioinformatics methods, ResultsA serotype Ⅰof DENV isolated strain named as SZ905 was identified to be genotype Ⅰ. And the homology analysis of E gene showed that the nucleotide homology with Malaysia 0802atw strain, Singapore strain, Zhejiang 17-45 strain, YN176282 strain and BNJH 17108 strain were all above 99%, and the homology of E protein with the multiple strains such as NJX-14, Zhejiang 17-45, Malaysia 0802atw and 1208atw strains was all above 99.8% in amino acid level, indicating that these strains maybe originate from a same original strain. And the evolution of E protein sequence is stable and highly homologous with that of several strains isolated in Guangdong Province since 2002. Conclusion DENV-Ⅰ genotype Ⅰ in China has stable genetic evolution and high degree of conservation, which is conducive to the prevention and control of this serotype virus disease in recent years.
