Expression of absent in melanoma 2 mRNA in peripheral blood mononuclears and its significance in primary hepatocellular carcinoma

摘要: 目的 探讨外周血单个核细胞黑色素瘤缺乏因子2(AIM2)在原发性肝癌(HCC)患者中的表达及其临床意义。方法 通过逆转录PCR和实时荧光定量PCR检测41例HCC患者外周血单个核细胞AIM2 mRNA的表达水平, 分析其与患者肝内肿瘤直径、肝外转移和肝功能储备评分的相互关系。结果肝内瘤体直径小于5cm的患者较肝内瘤体直径大于5cm的患者具有更高的AIM2 mRNA表达水平(P=0.006), 未发生肝外转移者较肝外转移者有更高的AIM2 mRNA表达水平(P=0.025), 肝功能储备A级和B级的患者较C级患者有更高的AIM2 mRNA表达水平(P<0.0001)。结论 外周血单个核细胞AIM2 mRNA的表达水平与HCC病情进展呈负相关, AIM2可能是HCC病情评估的潜在生物标志物和治疗干预的潜在靶点。

Abstract: Objective To analyze the expression level of absent in melanoma 2(AIM2) in peripheral blood mononuclears(PBMCs) and its significance in primary hepatocellular carcinoma(HCC). Methods The mRNA expression levels of AIM2 in PBMCs in 41 patients with HCC were detected by reverse transcription PCR and real-time fluorescence quantitative PCR, and the relationship between the liver tumor diameters, extrahepatic metastasis and liver function reserve scores were analyzed. Results Patients with the diameter of the intrahepatic tumor less than 5cm had higher AIM2 mRNA expression levels than those with the diameter of the intrahepatic tumor greater than 5cm (P = 0.006). The AIM2 mRNA expression levels were higher in patients without extrahepatic metastasis than in those with extrahepatic metastasis(P = 0.025). Patients with grade A and B liver function reserve had higher AIM2 mRNA expression levels than patients with grade C (P < 0.001). Conclusion The expression level of AIM2 mRNA in PBMCs is negatively associated with the progression of HCC. AIM2 is a potential biomarker for the prognostic evaluation of HCC and a potential target for therapeutic intervention.
