海绵状脑病致病朊蛋白对胆碱能神经元兴奋性和膜受体的影响

Effects of the pathogenic prion protein on neuronal excitability, membrane receptor in spongiform encephalopathy

摘要: 目的 探讨致病海绵状朊蛋白(PrP)肽段(PrP106-126)对大鼠基底前脑胆碱能神经元兴奋性和膜受体的影响。方法 采用全细胞膜片钳技术,记录急性分离的大鼠基底前脑胆碱神经元的动作电位发生频率。用AC253检查朊蛋白作用的细胞膜受体。结果 PrP106-126(10nmol/L)对基底前脑胆碱能神经元具有兴奋作用。在电流钳模式下,PrP106-126肽段诱发的动作电位频率增加,由 (2.2±1.9)Hz增加到(58.0±18.0)Hz(n=32,P<0.001)。使用胰淀素受体竞争性拮抗剂AC253(10nmol/L),可以阻断PrP106-126的作用。结论 PrP106-126对中枢神经元细胞膜兴奋性的影响,是通过胰淀素受体介导的。

Abstract: Objective To investigate the effects of the pathogenic prion protein (PrP) peptide (PrP106-126) on the excitability and membrane receptors of cholinergic neurons in rat basal forebrain.Methods Whole cell patch clamp technique was used to record the frequency of action potential on cholinergic neurons isolated from rat basal forebrain.The membrane receptor of PrP was detected by AC253.Results Our results showed that PrP106-126 (10 nmol/L) had excitatory effect on cholinergic neurons in basal forebrain.Under the model of current clamp,the frequency of action potential induced by PrP106-126 peptide increased from (2.2±1.9) Hz to (58.0±18.0) Hz (n=32,P<0.001).Using AC253 (10 nmol/L),a competitive antagonist of amylin receptor,can block the effect of PrP106-126.Conclusions The effect of PrP106-126 on the membrane excitability of central neurons is mediated by amylin receptor.
