Detection of Helicobacter pylori by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry

Abstract: Objective Establish the matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS)'s ability of identifying Helicobacter pylori (Hp) and evaluate the effect of Hp-resistant strains on the results. Methods The Hp strains which were verified by traditional methods, drug resistance tests and confirmed by full gene sequencing were used for building MALDI-TOF MS library. 10 drug-sensitive Hp strains from the library were randomly collected to establish Hp identification Super Spectrum. Then, 201 Hp strains obtained from newly collected gastric mucosal biopsy samples were tested and evaluated by the MALDI-TOF MS. They were verified by culture identification, drug resistance test, and complete gene sequencing as well. Results The 201 Hp strains were tested by the MALDI-TOF MS. 196 strains of them were positive. The positive rate of MALDI-TOF MS was 97.5% (196/201). 33 strains of them were drug sensitive, which were all positive and the positive rate was 100% (33/33); 168 stains were drug-resistant. 163 strains of them were positive and the positive rate was 97.0% (163/168). There was no statistically significant difference between the two groups (P>0.05). Conclusion The identification of Hp by MALDI-TOF MS is accurate, which can greatly shorten the time of culture and shows a good clinical application value. The Hp-resistant strains have no significant effect on the identification accuracy.
